Introduction of Chimeric IgA Antibody

IgA is the second most prevalent antibody in human serum and the most predominant antibody isotype in mucosal secretions, where it serves as the first line of defense and protects against pathogens that are ingested or inhaled. Studies have shown that IgA antibodies could effectively trigger respiratory burst and phagocytosis of various microbial pathogens by human leukocytes and be exceptionally effective in recruiting polymorphonuclear cells (PMN) for antibody-dependent cell-mediated cytotoxicity (ADCC) against tumor cells. Moreover, IgA has been reported to combat certain infectious and malignant diseases. Though numerous laboratory and clinical researches are being widely explored, IgA antibodies as therapeutics for human use still has a long way to go. Major drawbacks are that IgA forms in human serum are different from those in most animal species and the lack of an IgA receptor homolog in mice, which result in challenges in in vivo efficacy evaluation of therapeutic IgA antibodies. To overcome these shortcomings, multiple engineering approaches have been tried and utilized to improve the properties of recombinant monoclonal IgA antibodies, among which chimerism of IgA antibody is a typical modification.

Approaches for Chimeric IgA Antibody Production

The production of chimeric IgA antibodies usually adopts recombinant DNA technology due to the fusion of two species. Chimeric IgA antibodies production requires a sophisticated expression system capable of performing extensive post-translational modifications. Exactly, IgA production has been successfully demonstrated in our Expression System Platform, such as mammalian, insect, and plant cells. Especially, Chinese Hamster Ovary (CHO) cells are the most extensively applied host for recombinant IgA production. However, upstream processing of recombinant IgA seems to be in its infancy still, as the reported cultures are mostly conducted in simple batch mode and the working volumes are far from liter scale. During the past years, tremendous efforts have been devoted at Creative Biolabs to further optimize the applied medium formulation and develop more advanced process strategies to boost IgA titers and increase the process output to yield the desired amounts.
