Background of Antibody Expression Systems

Monoclonal antibodies play an increasingly critical role in the field of basic research, diagnostics, and treatment of human diseases since the first licensed monoclonal antibody Orthoclone OKT3 (muromonab-CD3) was approved in 1986. There are about 100 therapeutic antibodies have already been approved by the FDA and thousands of antibodies are in different stages of pre-clinical or clinical development up to now. Not surprisingly, the development of highly efficient systems for antibody expression or production is more and more important.

In terms of antibody production, the expression of non-IgG antibodies is different and more difficult than those IgG antibodies, mainly reflecting in (i) the number of glycosylation sites of IgA is much higher than IgG; (ii) IgM presents high molecule weight and genes involvement of light chain, heavy chain, and the J chain; (iii) extreme low abundance of IgE in healthy cells or tissues. As a senior expert in monoclonal antibodies production and development, Creative Biolabs has established a special and professional Non-IgG Antibody Expression Systems based on our seasoned IgG isotype monoclonal antibody technologies and platforms.

Features of Our Antibody Expression Systems

Firstly, we have constructed a full range of non-IgG antibody expressing cell lines. Several expression systems can be used for IgA antibody production, including Chinese hamster ovary (CHO) cell lines, plants, myeloma cells(NS0, SP2/0, YB2/0), baby hamster kidney cells, insect cells, and monkey-derived COS cells. Most of our recombinant IgA monoclonal antibodies are produced by CHO cell lines. We have successfully produced polymeric IgM by using rat hybridoma cell lines, PER. C6 human cell line, C6 glioma, HeLa cells, and CHO cells. And IgM production by rat hybridoma cells and PER. C6 cells are much higher than those in other cells. Secondly, glycosylation shows significant effects on antibodies like immunogenicity, half-life, bioactivity, etc., and non-human cells glycosylation may induce severe immunogenic reactions. Therefore, Creative Biolabs has intentionally established specific human glycol-engineered cells systems to provide better-humanized antibody products with optimized glycosylation.
