Introduction to IgE Antibody

IgE antibody is the least abundant circulating immunoglobulin isotype in mammal serum. IgE mediates various functions through binding to 2 Fcε receptors (FcεRs) mainly on the surface of mast cells and basophils, the high-affinity receptor FcεRI and the low-affinity receptor FcεRII (CD23). As an important member of humoral immunity, IgE triggers immune reactions for host defense to protect against parasitic worms (helminths) and eliminate environmental substances including toxins, venoms, irritants, and xenobiotics. Studies have shown that IgE antibodies play an essential role in inducing the allergic immune response (mainly type I hypersensitivity) when exposed to allergens, which results in mast cells and/or basophil degranulation, acute inflammation, and various allergic diseases. Furthermore, increasing evidence suggests that IgE may play a significant role in antitumor immunity. Especially, several chimeric IgE specific for certain cancer antigens recently have exhibited rather favorable anti-tumor effects.

Chimeric IgE Antibody Engineering

During the past years, though the IgE functions have been extensively investigated, the applications of IgE for clinical use are hampered as the expression level of nature IgE antibody is very low and it may elicit an unwanted immune response. In this case, IgE antibodies chimerized by replacing the antibody constant region with other species to reduce the antibody's immunogenicity while retaining the ability to specifically bind to the antigen have been widely used for antibody preparation. By this modification, chimeric IgE antibodies reduce anti-species immune responses when used in different animal models, particularly for human applications. For instance, mouse-human chimeric IgE antibody specific for the epithelial antigen MUC-1 (anti-MUC-1 IgE), mouse-human chimeric IgE against a prostate-specific antigen (anti-PSA IgE), anti-Ly-2 rat/human chimeric IgE, have been widely evaluated in IgE-based cancer therapies in vivo.
