Immunoglobulin E (IgE), a principle antibody involving anti-parasitic infection and allergic reaction, has been widely demonstrated to play an important role in evaluating patients with IgE-mediated disorders, allergic diseases as well as cancers. Actually, the concentration, binding ability, and the specificity of IgE antibodies all can affect the humoral immune response in disease progression. Firstly and particularly, an increased affinity of IgE antibody to allergen and an increased concentration of specific IgE antibody or total serum IgE can eventually cause a high level of basophil and mast cells of patients, by which these specific IgE antibodies and IgE-related cells are beneficial tools for allergic potential risk identification, allergic diseases diagnosis, and clinical symptoms monitor. Secondly and importantly, the development of IgE-based therapies targeting different types of cancer antigens have been evaluated in a number of studies, and the results have demonstrated that IgE also is a powerful diagnostic tool for the diversity of cancers. Therefore, a variety of methods have been established for testing IgE, including in vivo skin test, various serological methods, and clinical testing.

The Assays for lgE Diagnosis

Radioallergosorbent test

Radioallergosorbent test, also known as RAST, is one of the most common methods for detecting specific IgE antibodies, by which, it can test the potential or known allergens and provide data for allergic disease diagnosis. In general, adding the test allergen into the serum samples of patients, antibodies can closely bind to the allergen if the serum has positive IgE antibodies. Radially labeled anti-human IgE antibodies can interact with the IgE antibodies- allergen complex, and unbound anti-human IgE antibodies will be washed away by buffers in this system. The amount of radiation is positively correlated to the allergen serum IgE, and thereby the concentration could be further analyzed and calculated.

The nonisotopic auto-analyzers assay

Nonisotopic auto-analyzers assay, a method basing on RAST, is widely used to quantify IgE antibodies all over the world. In this assay, allergen-specific IgE antibodies are cultured into solid-phase allergens and then labeled by anti-human IgE antibodies. In addition, the signal of radiation can be captured by the device, and a total serum IgE calibration curve will be formed which plays an important role in quantifying IgE antibodies in this system.
