IgM, also known as immunoglobulin M, is the largest antibody that is produced by the immune system of vertebrates. It can attack against a wide variety of antigens and take part in many steps of the primary and adaptive humoral immune response. It is noteworthy that IgM can more effectively activate the complement system than any other isotypes, contributing to a number of cytotoxic and cytolytic reactions. In structural, the IgM monomer consists of two identical heavy chains (μ) and two light chains (κ or λ), and each chain can be divided into two domains, a variable (V) domain, and four different constant (C) domains. The variable domain of IgM is usually used for binding to antigens, and then the constant domains of IgM can interact with phagocytic cells and the complement system. Meanwhile, recent studies have revealed that recombinant IgM antibodies can be regarded as a therapeutic agent in cancer immunotherapy and has been performed related to a number of carcinomas, such as epithelial ovarian cancer (EOC), breast cancer, as well as lung cancer.
